the original Xbox Live servers were discontinued 12 years ago so no, it's not possible. DOA2U is only playable online via insignia on a modded Xbox.

Should clarify the Xbox doesn't have to be modded to connect to Insignia.

That said, there's no reason for you to not mod your Xbox at this point.
